RESUMO

Understanding the molecular composition and fate of dissolved organic matter (DOM) during transport in estuaries is essential for gaining a comprehensive understanding of its role within the global biogeochemical cycle. In 2020, a catastrophic flood occurred in the Yangtze River basin. It is currently unknown whether differences in hydrologic conditions due to extreme flooding will significantly impact the estuarine to oceanic DOM cycle. We determined the DOM composition in the Yangtze River estuary (YRE) to the East China Sea by using Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) during the high discharge and the flood period (monthly average discharge was 1.2 times higher) on the same trajectory. Our study found that the composition of DOM is more diverse, and more DOM molecules were introduced to the YRE during the flood, especially in the freshwater end member. The result revealed that the DOM was significantly labile and unstable during the flood period. A total of 1840 unique molecular formulas were identified during the flood period, most of which were CHON, CHONS, and CHOS compounds, most likely resulting from anthropogenic inputs from upstream. Only 194 of these molecules were detected in the seawater end member after transporting to the sea, suggesting that the YRE served as a 'filter' of DOM. However, the flood enhances the transport of a group of terrigenous DOM, that is resistant to photodegradation and biodegradation. As a result, YRE experienced ~1.6 times higher terrigenous DOC flux than high discharge period. Considering the increased frequency of future floods, our study provides a preliminary basis for further research on how floods affect the composition and characteristics of estuarine DOM. With the help of the FT-ICR MS technique, we can now better understand the dynamic of DOM composition and characteristics in large river estuaries.

Analyzing the molecular composition change of dissolved organic matter (DOM) during transportation in estuaries can enhance our understanding of the fate of DOM. However, the impact of hydrologic conditions resulting from large river plumes on the DOM cycle are less explored, and previous studies were insufficient to capture the molecular fate that occur during the transportation process. In this study, we used a range of bulk and optical techniques, as well as Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), to determine the concentration and characteristics of DOM along two trajectories of downstream plumes of diluted water of the Yangtze (Changjiang) River estuary (YRE) during the high discharge season. These two plumes situated along the route of the summer Changjiang diluted water (CDW) have been identified and named CDW-North (CDW-N) and CDW-South (CDW-S), respectively. Despite having the same riverine end-member origin, the turbidity zone in YRE significantly modifies the molecular characteristics and composition of DOM. The results of FT-ICR MS indicated a spatial variation of DOM composition in the coastal zone of the two plumes. The relative intensities of the CHO, CHOS, and CHONS compounds are negatively correlated with salinity. In addition, the coastal zones of both CDW-N and CDW-S are characterized by more autochthonous DOM sources. More CHON compounds in CDW-N are probably due to the production of autochthonous DOM in offshore waters. The activity of phytoplankton increased the surface dissolved oxygen level of CDW-N in the coastal zone. However, the hypoxic zone formed at the bottom of the CDW-N due to microbial degradation of organic matter and may further benefit the preservation of CHON compounds. Our study emphasizes that the characteristics and composition of the estuarine DOM can be significantly shaped by distinct large river plumes. Furthermore, using FT-ICR MS in combination with complementary techniques can better assist in identifying the sources and transformation mechanisms of estuarine DOM in large river plume-affected systems and provide more valuable insights into the role of DOM in the estuarine biogeochemical cycle.

We investigated the sediment carbon (C) stocks, sequestration and sources in tidal flats and their adjacent mangroves in two coastal wetlands in Hong Kong (the Mai Po Nature Reserve (MPNR) and Ting Kok (TK)), part of a megacity of ∼20 million. At both locations, the C stock of tidal flats was lower than that of mangroves. In MPNR, tidal flats indicated a higher C burial rate (75.2 g C m-2 yr-1) compared to mangroves (64.3 g C m-2 yr-1). The C of tidal flats mainly originated from mangroves and riverine sources, while those in deeper sediments (below 60 cm) in the mangroves originated from the tidal flats. In TK, the C of tidal flats was dominated by oceanic sources. Our study indicates that tidal flats are also important in coastal C sequestration. On highly urbanized coasts, tidal flats and their connected mangroves can play a critical role for C mitigation.

@#To investigate the current status of online classes, screen time and its influencing factors among primary school students in Guangdong during the 2019 novel coronavirus pandemic.@*Methods@#Using the convenience sampling method, a total of 5 266 pupils aged 6-12-years-old from Guangzhou, Zhanjiang, and Zhongshan participated in the online questionnaire. ANOVA or chi square tests were performed to compare differences in online classes and screen time between grades, and multinomial Logistic regression was performed to analyze the correlates of recreational screen time.@*Results@#The prevalence of prolonged recreational screen time was 42.2% and 55.2% on weekdays and weekends, respectively. Recreational screen time increased by 40.31 min/d on weekdays and 33.07 min/d on weekends, compared to usual school semesters. The average duration of an online class was (26.07±9.62) min, which totaled (110.41±51.98)min per day. Sex, grade, being the only child, and parents education levels were identified as the influencing factors of prolonged recreational screen time. Children who practiced moderate levels (weekdays: OR =1.27; weekends: OR =1.40; P <0.05) or lower levels of physical activity (weekdays: OR =1.86; weekend: OR =1.84; P < 0.05 ) were at a higher risk of prolonged recreational screen time than those who practiced more vigorous physical activity. Children whose parents limited their screen time to a moderate (weekdays: OR=1.61, P <0.05) or lower level (weekdays: OR=1.32, P < 0.05 ) had a higher risk of prolonged recreational screen time than those with a higher frequency. Children with parents recreational screen time ≥ 2 h/d had a higher risk of prolonged recreational screen time than the reference group; children who exhibited moderate to vigorous levels of physical activity <1 h/d (weekdays: OR=1.31, P <0.05), and those used electronic devices for learning 1-2 h/d (weekdays: OR =2.65; weekend: OR =2.65; P <0.05) or for ≥2 h/d (weekdays: OR =4.05, weekend: OR=5.24, P < 0.05 ) were at a higher risk of prolonged recreational screen time than the reference group.@*Conclusion@#During the COVID-19 pandemic, the level of screen time among children in Guangdong was high. Behavioral monitoring and targeted interventions are needed to promote children s health.

The gene expression chip of a salt-tolerant wheat mutant under salt stress was used to clone a salt-induced gene with unknown functions. This gene was designated as TaSR (Triticum aestivum salt-response gene) and submitted to GenBank under accession number EF580107. Quantitative polymerase chain reaction (PCR) analysis showed that gene expression was induced by salt stress. Arabidopsis and rice (Oryza sativa) plants expressing TaSR presented higher salt tolerance than the controls, whereas AtSR mutant and RNA interference rice plants were more sensitive to salt. Under salt stress, TaSR reduced Na(+) concentration and improved cellular K(+) and Ca(2+) concentrations; this gene was also localized on the cell membrane. ß-Glucuronidase (GUS) staining and GUS fluorescence quantitative determination were conducted through fragmentation cloning of the TaSR promoter. Salt stress-responsive elements were detected at 588-1074 bp upstream of the start codon. GUS quantitative tests of the full-length promoter in different tissues indicated that promoter activity was highest in the leaf under salt stress. Bimolecular fluorescence complementation and yeast two-hybrid screening further showed the correlation of TaSR with TaPRK and TaKPP. In vitro phosphorylation of TaSR and TaPRK2697 showed that TaPRK2697 did not phosphorylate TaSR. This study revealed that the novel TaSR may be used to improve plant tolerance to salt stress.

The study with impregnating method showed that after overwintering, the susceptibility of Carposina niponensis larvae on triazophos, phoxim, chlorpyrifos, malathion, lambda-cyhalothrin and abamectin was 34.50, 16.71, 3.89, 3.28, 5.90 and 2.73 times as much as that before overwintering, the total protein, glycogen and fat contents and carboxylesterase, acid phosphatase, alkaline phosphatase, glutathione-S-transferase, superoxide dismutase, catalase and peroxidase activities in C. niponensis larvae were decreased by 17.10%, 41.76% and 30.08%, 62.36%, 53.47%, 76.19%, 80.60%, 18.77%, 14.16% and 64.02%, respectively, and the activity of acetylcholinesterase, the target enzyme of many insecticides, was 1.41 times as much as that before overwintering. It was suggested that the difference of the susceptibility was resulted from the changes in the contents of biochemical substances and the activities of metabolic enzymes, protective enzymes and target enzyme.
